The role of peroxidase in the bactericidal activity of human blood eosinophils.

چکیده
Utilizing cell preparations that contained greater than 90% eosinophils and sodium azide to inhibit the lysosomal enzyme peroxidase, studies of eosinophil bactericidal activity were undertaken. Eosinophils did not kill bacteria as well as neutrophils, principally because of a reduced phagocytic capacity. In contrast to neutrophils, eosinophils had high resting iodination activity but failed to increase this activity following phagocytosis. One millimolar sodium azide, which inhibits both eosinophil and neutrophil peroxidase as measured by the iodination of tnchloroacetic acid precipitable protein, impaired neutrophil staphylocidal activity, but enhanced eosinophil killing of Staphylococcus aureus primarily by increasing phagocytic uptake. Bactericidal studies utilizing lysostaphin suggest that neutrophil peroxidase exerts its contribution to bactericidal activity only during the early postphagocytic period. Eosinophil peroxidase is genetically and biochemically distinct from neutrophil peroxidase, and appears to play no role in the bactericidal activity of intact eosinophils.

Human eosinophilic peroxidase: role in bactericidal activity.
Peroxidase, present in both neutrophils and eosinophils, is thought to be involved in the bactericidal activity of leukocytes, through either the decarboxylation of amino acids in the presence of chloride or the iodination of proteins in the presence of iodide. We examined these activities in sonic extracts of purified neutrophils and eosinophils and related them to the ability of the extracts ...

Eosinophils from a patient with hypereosinophilia were observed to phagocytize radiolabeled E. coli or S. aureus as effectively as neutrophils from a normal control. This was observed at a number of bacteria/cell ratios and a various time intervals following initial challenge and was confirmed by direct microscopic examination. In spite of comparables rates of phagocytosis, the eosinophils were...

Eosinophils participate in allergic inflammation and may have roles in the body's defense against helminthic infestation. Even under noninflammatory conditions, eosinophils are present in the mucosa of the large intestine, where large numbers of gram-negative bacteria reside. Therefore, roles for eosinophils in host defenses against bacterial invasion are possible. In a system for bacterial via...

Eosinophils participate in the inflammatory response seen in allergy and parasitic infestation, but a role in host defense against bacterial infection is not settled. The bactericidal/permeability-increasing protein (BPI) has been demonstrated in neutrophils and it exerts bacteriostatic and bactericidal effects against a wide variety of Gram-negative bacterial species. Using the Western blot te...
متن کاملEVALUATION OF GLUTATHIONE PEROXIDE ACTIVITY AND OXIDATIVE STRESS IN TYPE 2 DIABETIC PATIENTS AND THEIR RELATIONSHIP WITH SERUM GLUCOSE LEVEL AND BLOOD LIPID PARAMETERS IN A CASE-CONTROL STUDY
Background & Aims: Diabetes has been the most common metabolic disorder in recent decades. Oxidative stress and its related factors play an important role in the onset and complications of diabetes. In this study, glutathione peroxidase activity and oxidative stress indices in serum of type 2 diabetes patients and their correlation with blood glucose and lipid parameters were evaluated. Materi...
